Barbara- You might try the ccf2 substrate system reported in Jan 1998 Science (Zlokarnik et al). It does not require hypotonic shock to load, and the emission wavelength is dependent upon clevage by beta-gal, giving an internal control for loading. It has been used in our facility with good success. It excites optimally in the low 400 nm range, ideal for a violet-enhanced krypton laser, but may work as well with the uv lines of an argon laser. -Marty Bigos Stanford Shared FACS Facility
