Hi Folks, My first posting. I'm pretty new to FACS (most of my experience is in enzyme assays and molecular biology). What I'm trying to do right now is smash up some PBMCs to generate debris for use as a standard to characterize naturally occurring debris from expansion cultures. I tried freeze-thaw but got only aggregated material--I need a method that solubilizes everything, so that I can FACS the debris from a defined number of cells. What do people think?
