I am working with an investigator that would like to evaluate monocyte progenitors in mouse bone marrow. He is specifically interested in identifying progenitors that have already committed to the myeloid/macrophage lineage and pro-monocytes. In our preliminary experiments, we have had trouble distinguishing a recognizable pattern of marker expression to isolate these small populations. We have been using CD34 (RAM34), CD31(390), and Ly-6C(AL-21) antibodies, as per several references we found. Has anyone else had success using this combination? Is there an alternate staining strategy that someone could recommend for this purpose? My investigator and I are both new to this type of analysis, so any insight or advice (no matter how elementary) would be very much appreciated. Thank you!
