I'm using the ISHAGE single-platform technique through use of Beckman-Coulter's Stem
Kit. I use a stop count of 100K for CD45+ events and microspheres or a stop time of 10
minutes. After 10 minutes, the microspheres either float or sediment, requiring that
the acquisition be paused, the tube removed to be vortexed and the sample acquisition
resumed.
Ideally, 100 events are analyzed.

Dear all,

We usually acquired 75,000 CD45+ events for CD34 enumerations. We do not
process the test if cell count is less than 1000/ul but there are occasions
where 75,000 collection is not possible. I am interested to know what is
the usual practice in other clinical labs when this happen? What is the
minimum acceptable number of events collected to report CD34 count?

Thank you.

Ching Ying
