while we are discussing apoptosis scatter of T cells. I have a question
regarding scatter of apoptotic neutrophils. I have attached a file showing
the scatter of neutrophils in fresh blood and whole blood incubated with
TNFa for 24h to induce apoptosis. CD16 shedding was used as the criteria to
monitor apoptotic neutrophils. What I don't understand is that I look at
CD16 staining on cells in Gate R1. When I set up a gate on Cd16-lo cells the
pink population comes up. If I look at CD16 on cells in the pink population
(low side and forward scatter) in the 24h incubated cells, the CD16 staining
is coinciding with apoptotic peak (CD16lo).

Can somebody please tell what can I conclude out of this. Also does it mean
that we have to take into account the cells with a low side and forward
scatter. Usually I gate on the CD16-lo population and look at that
population for further events (like caspase 3 etc)

I have attached a MAC file as well as a PC file so that both users can open
it.
