Hello, Need some help on determining cytoplasmic Kappa/Lambda ratio for plasma cells. Can not decide which antibody combination is the best. Option #1 Tube 1: Isotope K/L + CD45 Tube 2: CD38/CD56/CD/45 Tube 3: Kappa/Lambda/CD45 Gate on CD38 bright and CD56 positive population (plasma cells) and determine K/L ratio Option #2 Tube 1: Isotope K/L + CD45 Tube 2: Kappa/CD38/CD45 Tube 3: Lambda/CD38/CD45 Gate on CD38 bright (>10E3 log) and look at K & L fluorescence intensity difference. How to determine the K/L ratio? May be neither of them are the best combination. Any helpful suggestion will be appreciated. Looking forward to hearing from people soon.
