Dear flowers, We're about to perform bcl-2 detection in PBMC, HeLa and K562 as a part of previously designed experimental conditions, by flow cytometry. What would be the most appropriate method for intracellular bcl-2 detection with FACS? Is it possible to follow BD permeabilization procedure designed for intracellular cytokine staining (without stimulation, of course)? Could anyone suggest something special, experienced with Sigma's anti-bcl-2 mAb and what would be the expected bcl-2 staining values for above mentioned nontreated cell suspensions?
