You might consider trying to detect antigens expressed by proliferating cells. For instance, antibodies to Ki-67 - an intracellular antigen expressed in dividing cells - are available and can be used with surface antibodies like CD3, -4, or -8. Such a method would allow you to quantify the percentage of dividing T cells in your sample.
