Howard, basically keep it as it is. The 3rd edition is a very good and comprehensive information source of all aspects of flow cytometry, and in my view it should stay that way. To my knowledge there are quite a few books about practical applications, and that will always come down to something like cook books for specialized users. However, one thing that could be worth considerung might be an additional chapter that focuses on practical details that you don´t find in the manual (to quote yourself: "...the practical wisdom doesn´t always get written down..."), and that are relevant to users in all fields (something like the paper by Phil Dean "Helpful hints ...", but covering all other aspects of the technical side of flow cytometry).
