Dear Lee,

We developed a very fast-automated whole blood lysing technique based on an
inorganic-acid.
In this method lysing of the red cells is established by a very short
pH-shock. 
A disadvantage of this technique is that we have not been able to use a
buffer like PBS for the dilution of whole blood.
We tested a lot of alternatives and found very good results with Alsevers
solution (Sigma, product number A-3551, contains 4.2g NaCl, 8.0g sodium
citrate and 20.5g glucose/L).
When diluting in this solution we have even seen an improvement of the
morphology and antigenicity over the not diluted whole blood.
