Subject: CD34 cell enumerations

>Can anyone recommend protocols and reagents for enumeration of human CD34+
>cells in cord blood, also in cytokine/factor stimulated cultured leucocytes
from
>different sources. I am using a BD FACS Sort with a Mac (CellQuest software),
>and am a newcomer to this setup and FACS in general. In preliminary
experiments
>I have used BD RPE-anti-CD34 and BD FITC-anti-CD45 - still playing around with
>settings - lots of questions, but getting fascinated. Is CD45 an optimal
marker
>for total leucocyte detection - should I be going for a panel of
>lineage-specific markers - any recommendations and advice on this would be
>appreciated.

You may get some ideas and references from a paper we published in the
latest issue of Exp Haematol [Differential effects of the hematopoietic
inhibitors MIP1a, TGFb, and TNFa on cytokine-induced proliferation of
subpopulations of CD34+ cells purified from cord blood and fetal liver.
23:422-427, 1995 Mayani et al. ] We used CD34, CD45RA and CD71.
