We have extensive experience with CLL B cells in a variety of settings. At
present we are using the B-D kappa/lambda for light chain determinations.
This works very well and you can combine it with a third color CD19 to
identify the clonal population. The most important thing to remember, is to
do light chain staining on PBMC (ficolled cells) and not whole blood.
Lysing rbc and washing before hand also works, but not as well. You can
take advantage of the fact that only one light chain will be expressed to
determine the phenotype even when the expression is quite dim. We have
also used Immunotech's k/CD19 and l/CD19 which also work.
