Dear Mike:
A very good manuscript on the subject is: Graham, J.K., Kunze, E and
Hammerstedt, RH, Analysis of sperm cell viability, acrosomal integrity, and
mitochondrial function using flow cytometry. Biol. of Reproduction 43:55-64.
Acrosome intact cells do not bind labeled (FITC or other dye) Pisum sativum
agglutinin (PSA) in the acrosomal region, while acrosome-reacted cells bind
PSA. Hope this is of some help.

I have a user who is looking for a dye or dyes that are specific for the
sperm acrosome. Has anyone out there been doing work in this area, and have
some suggestions for possible dyes? Thanks for any input.
