Second, we are exploring our options for re-configuring our clinical
operation (as opposed to our research lab) and have begun looking at the
Coulter XL with the autoloader. We have liked what we have seen so far but
some sites have talked about having difficulty setting fluorescence
compensation properly so that it works on all samples - this is said to be a
possible problem for simple FITC/PE as well as more complex arrangements. I
have not tried this myself yet but I am wondering if anyone else has seen a
problem and, if so, if you know its etiology. We did note that some samples
we gave to another site to look at with an XL had what appeared to be some
problems in this realm although I initially attributed it to a few "weak"
MoAbs coupled with setting comps based only on beads. Any comments (or any
general comments on the Coulter) would be greatly appreciated.
