REGARDING Protease digestion

Steve-
I have been having a difficult time with background when I use my .221 LCL
cells (lymphoblastoid cell line irradiated to not express MHC antigens). I
found a paper by Peter Lobo et al. (Transplantation 23:16, 1977) in which
they use protease to decrease background when using B cells for flow
cytometry. The pronase he used is no longer available, and he is out of the
country for a while. I tried using a similar protease, but it destroyed my
cells. Do you have any experience with this or do you know anyone that does?

My address is lynn_westphal@quickmail.ucsf.edu
I really appreciate any advice you have.
