[Preservative vs Stability]

We are doing some immunophenotyping with cyno monkey blood. We are
evaluating the stability of the blood at room temperature and using Streck
stabilizer (stored at 4C). All of our cell populations remain stable except
we see diminished B cells as early as 24 hours when samples are collected in
EDTA or heparin tubes. Has anyone else had similar issues? We are still
evaluating our stabilized samples but with our results to  date we have an
occasional animal where the B cells are much lower after 24 or more hours in
stabilizer than when we processed the blood immediately. I know for human
work folks use samples collected in heparin and I have not seen reports of
decreased B cells when samples are tested 24 hours after collection. If
anyone else has had similar issues I would be interested in hearing from
you.
