Hello!

I am an absolute beginner in the FACS (and immunology) business and I am
looking for advice on interpreting my FACS data. I have stained spleen and
tumor cells using two antibody mixes:
1) CD4, CD8, CD3, NK
2) MHCI, MHCII, B7-1
The animals had been treated with an immunostimulating adenovirus
intratumorally two weeks before sacrifice.
I would like to know the relative numbers of CD4, CD8, NK, MHCI, MHCII and
B7-1 positive cells compared to the control. As a control I only have
samples from untreated animals (mock adenovirus). So far I know that cells
double positive for CD4 and CD3 or for CD8 and CD3 can be counted as
positive for CD4 or CD8 respectively. The double positive cells would be
the upper right cell population. Unfortunately I don't know how to evaluate
the rest of the data and I don't have anybody around who knows. Any advice
on how to extract some information from such data as well as how to better
design such staining experiments in the future is highly appreciated.
Advice on where to get such information elsewhere or whether my question
should be better posted in another forum is also welcome. By the way, the
animals were not only sacrificed for the FACS analysis but managing this
would make the other data much more valueable.
Thanks for your help!

With kind regards,
Reinhard
