Hi,
I've never gotten good CVs when staining yeast DNA with PI. A former
colleague of mine had much better luck using mithramycin A, but you need a
457 line on your laser to get good excitation.	For a brief protocol see:

Frykman S, Srienc F. Cell cycle-dependent protein secretion by
Saccharomyces cerevisiae. Biotechnol Bioeng. 2001 Nov;76(3):259-68.

Subject: Yeast DNA staining

Hi Everyone,
I am looking for a good procedure for staining yeast DNA. I have
tried an
ethanol fix/PI procedure but the CV were in the teens. I plan to try
using
	SYBR Green next week.
Best regards, jsh
