Greg, I used intracellular histamine detection by flow cytometry. The cells were fixed in a mixture of 4% paraformaldehide and 4% carbodiimide, permeabilized with saponine. I used antibody to histamine (developed in rabbit, Sigma: Anti-histamine, cat. No: H 7403) and FITC-conjugated anti-rabbit antibody (BD) and this worked all right. I worked on macrophages and dendritic cells differentiated from monocytes in vitro, the results can be found in: Immunology Letters 76 (2001):175-182.
