Hi Phil...

Although a 488 nm line will work for DsRed, it prefers a green or yellow line.  You can tune most water-cooled (and some air-cooled) argon lasers to the 514.5 for enhanced excitation of DsRed, DsRed2 and HcRed.  If you have access to a mixed gas or krypton laser, the 520, 530 and 568 nm lines from these sources work even better (particularly for HcRed).  If you use the 568 nm yellow line, make sure your DsRed bandpass filter is blocked for 568 nm light (or use a 568 nm restriction bandpass along with your filter).

We (in collaboration with Teresa and Bob Hawley at the ARC Holland Lab) have some comparative excitation data for DsRed at all of the above wavelengths posted on our website (http://home.ncifcrf.gov/ccr/flowcore/DsRed.htm).
