Thanks Bye Simona We have used the DX5 from Pharmingen on spleen cells from mice. Of total lymphocytes, it routinely showed only 3 - 5% NK cells. Have you titrated the DX5? We use 500 ng per million spleen cells and allow it to stain for 30 minutes on wet ice. What blocking agent(s) are you using? We use PBS with 0.2% BSA and 0.02% sodium azide.
