This area has been extensively covered by our group and can be found in several publications. A good place to start would be: Keeney M, Chin-Yee IH, Weir K, Popma J, Nayar R, Sutherland DR. Single Platform Flow Cytometric Absolute CD34+ Cell Counts Based On The ISHAGE Guidelines. Cytometry (Communications in Clinical Cytometry) 34:61-70, 1998. Gratama JW, Keeney M, and Sutherland DR. Enumeration of CD34+ Hematopoietic Stem and Progenitor Cells. Current Protocols in Cytometry (Suppl 8), 6.4.2-6.4.22, 1999 For an extensive discussion on CD34 issues check out the following website: http://www.ishage.org/committees/Committees/Graft_Evaluation/graft.htm Rob Sutherland and I have developed a Frequently Asked Questions section here for the ISCT graft evaluation committee and also have listmode files and templates for both BD and Coulter instrumentation. In addition, Beckman Coulter has a kit (StemKIT) which is based on the single platform ISHAGE guidelines with 7-AAD and includes information on the correct quality control procedures required for this analysis. This kit works equally as well with BD or Coulter instrumentation. Feel free to contact either myself or Rob Sutherland (rob.sutherland@utoronto.ca) if you would need further information. These guidelines will be for users not familiar with doing this type of analysis. Any suggestions will be appreciated. Thanks, Jim Kurtz James W.
