Hi Nadeem,
   We routinely do CSF testing for tumor. The volume we receive is usually
3-5cc. I don't know if we have ever received a normal as most of the
patients have AIDS or LYG and the MD is looking to see if a secondary
malignancy has developed. We spin the specimen down and decant all but about
50ul. I use 6.5ul times 2 to look at viability and do a cell count with
crystal violet. If any RBC are seen in the trypan blue tube, it is noted and
put in the report.
The most cells I have counted was 300,000 which means that you have to have
a fair idea of what to look for first.

Can anyone point me in the right direction for immunophenotyping cells from
cerebrospinal fluid.  Specific questions I have are:

1.  What kind of volume is usually taken
2.  What are the normal population of cells - phenotype, nos. ?
3.  Any safeguards/controls for blood contamination ?

I should point out that we are looking to establish techniques to look at
cellular infiltrates (if we can isolate any) in CSF of macaques infected
with SIV/SHIV that subsequently develop NeuroAIDS.  Any advice, pointers to
papers would be most welcome human or macaque work.
