Kevyn Adler, One of our clients wanted us to conduct endotoxin-free sorts on a FACS Vantage. Our procedure entailed preparing the cytometer with the following run sequentially through both the sheath tank and the sample port: 1% ES 7X, 10% bleach, 70% ethanol, distilled water, and finally "blood bank" quality saline. We received no indication from the client that endotoxins were ever present in the sorted samples, although I do not know if the samples were tested for endotoxin. After cleaning your FACScan, I would recommend installing a new sheath filter before the final saline wash. We use saline from Bio Test Medical, available from Fisher Scientific, catalog number MSC1000B.
