Remember what you are asking the Pall filter to do. That is to hold back bacteria for a significant period of time. Most filters are composed of a membrane with holes in it. The holes are roughly all the same size. ie .22 micron. When you increase the pressure, these holes may be altered in their shape and allow for bacterial or yeast to pass the filter. It is a good idea to replace the filter often. Remember, the time to develop a cell line or sort product is a lot more costly than changing the fitler. We keep several filters around and autoclave them regularly. This way you can change them weekly or even daily if you choose.
