No matter what you read it is impossible to do Cyclin D1 by flow with available antibodies. We tried and tried (despite having been told it doesn't work by another very well known flow person) and found that the antibody picks up something else that must be destroyed by fixation and paraffin embedding. Since then I have heard from other excellent labs that they tried and tried and couldn't do it. You can get staining in mantle cell lymphoma but you can also get it in CLL- garden variety CLL. Now I could rush out and write another bad flow paper about cyclin D1 in CLL that can only be detected by flow (negative on sections) but I know that is bogus. It is not specific by flow and a non-specific cyclin D1 is worthless.
