One of my colleges is working with IC cytokine staining for IL-12 and TNFa in PBMC monocyte-depleted . They are having too much problems to identify cytokine secreting cells. a) They need a positive control for IL-12 and TNFa in lymphocytes. b) There is a mitogen or antigen that stimulate these cytokine secretion in lymphocytes. c) What are the relative percentage of IL-12 and TNFa secreting lymphocytes in peripheral blood.
