John-

BD sells a positive control for IC cytokine staining. They're called
HiCK cells, numbers 1-4, depending on which cytokine you're interested
in controlling for.

HiCK-1 is the one you want for TNF-a and HiCK-3 is for IL-12.

One of my colleges is working with IC cytokine staining for IL-12 and
TNFa in PBMC monocyte-depleted . They are having too much problems to
identify cytokine secreting cells.

a) They need a positive control for IL-12 and TNFa in lymphocytes.

b) There is a mitogen or antigen that stimulate these cytokine secretion
in lymphocytes.

c) What are the relative percentage of IL-12 and TNFa secreting
lymphocytes in peripheral blood.
