just a comment on annexin V staining. We have used it routinely to detect apoptosis induced by toxins and it generally correlates well with other apototic indices (dna frag etc). However I would mention a paper we published in 1999 (Cell Death & Diff. vol 6 p624-637) which describes PS exposure as assessed by annexin V - FITC which occurs in PI negative thymocytes which go on to die exclusively by necrosis. We have not looked at any other cell types in detail but suspect that some cellular insults which cause a rise in calcium in the cell and result in necrotic cell death may also cause PS exposure prior to overt membrane damage (ie no PI uptake) So beware of using PS exposure as your only measure of apoptotic cell death Paul Waring Chemistry Department ANU, Australia
