Hello.

I am not currently in the mailing list, but I have received a forwarded
message regarding the question on Annexin V for analyzing apoptosis.  It
seems that she might have gotten answers to her questions already, but I
will give a short response since I have worked with it.

Annexin staining to detect early apoptotic process using a flow cytometry
is quite straight forward and simple.  Annexin V comes as FITC- or
PE-conjugated, so that you can pick and choose depending on what color your
antibody is conjugated with if you are double staining.  If FITC conjugated
Annexin is used, of course as all of you know, you can quantify the %
apoptotic cells by gating on FL1 positive populations.   For PE, FL2
positive, etc.

If you have any further questions regarding gating, cytosettings (e.g.,
voltage, compensation, etc), etc, please don't hesitate to contact me.  I
am still researching and learning about various assays to detect and
quantify apoptosis using flow cytometry myself, but I will be happy to
share what I know about Annexin V staining.
