Tina--note that proper compensation does not require the whole negative population to be on-scale: it only requires that the MEDIAN of the negative population be on-scale. If you are not used to running PBMC, you may be surprised by the relatively low autofluorescence that they have, making it indeed impossible, at times, to push all cells of the axis. Just set the voltage so that the top of the negative population is near the top of the first decade--you should be OK then. mr
