Recently, I was reading an article where the investigators subtracted the mean fluorescence intensity of the negative (isotype)control population from the fluorescence intensity of the positively stained cells.  I was wondering what folks thought of this practice.  Is this legitimate? and if so (or not) under what circumstances?
