Hi, the expression of light chain on CLL is very low. We use also the whole blood assay but usually wash the cells before staining to remove serum. If your assay is well standardized in terms of the same fluorescence intensity for normal kappa and lambda cells, you can compare fluorescence intensity of kappa and lambda by Kolmogorov-Smirnov analysis.
