I guess that the extinction coefficient of avidin should be known. You can also rapidly measure it, if you have pure protein. the sequence of avidin is also known. programs like PEPTIDESORT of GCG calculate the extinction coefficient from the sequence (content of W and Y, with minor influence of C and F). we have tested it in a couple of occasions and it is, indeed, fairly accurate.
