Look at

Baumgarth, N., Egerton, M. & Kelso, A.  Activated T cells from draining lymph
nodes and an effector site differ in their responses to TCR stimulation. J.
Immunol. 159:1182-91 (1997).

This paper didn't do ELISA, but did do bio-assays.  In addition, we have done
extensive comparison of cytokine profiles by mRNA and by FACS in human T cell
subsets, finding good correlation--hopefully will be published very soon.  In
addition to the frequency of producers being a correlate of production by the
whole population, the relative fluorescence within cells appears to correlate
(grossly) with their production.  However, I wouldn't rely on this latter
correlation for anything better than order-of-magnitude.
