I am looking for expertise for performing heterokaryon analysis by FACS. The overall goal is to examine the effect of cell fusion on the conformation of a cell surface molecule, 'X'. Cell types 'A' and 'B' can be uniquely distinguished by antibodies to cell surface molecules, and the conformation of X can also be distinguished by reactivity with a panel of antibodies. I would think one could fuse A with B using PEG, then identify heterokaryons and determine the conformation of X all using FACS. Anyone know of any references to this type of experiment, or personal experience that can help me think through the potential problems with analysis?
