Hi All, I am trying to figure out the laser spot intensity used by some investigators in a paper, an effort hampered by the fact that some of the details are missing from their methods. Ultimately, I need to determine an approximate intensity of the excitation beam for the following two instruments: (1) a laser-based "FCS" made by Jasco in Tokyo (400 mW at 488 nm) (2) a FACS Analyzer by BD (mercury-cadmium arc lamp at 485 nm) For the FCS system, I need to figure out if there is a "standard" optics configuration so that I can determine the spot size, and for the FACS Analyzer, I need both an estimate of the lamp intensity and a description of the optical configuration. Any help would be greatly appreciated! Kent
