     You might want to try a Stomacher (Tekmar-Dohrmann; Cincinnati, OH 
     513/247-7000)for your plant tissues. I was able to get suitable 
     samples from potato leaf tissue for DNA analysis, although it was only 
     a couple of experiments.

Hi,
I have recently obtained a Dako medimachine to isolate nuclei 
from plant leaf tissue.  Right now staring me in the face are 500 
hops and 1200 kiwifruit samples and all I am getting is very high 
levels of debris and no distinct nuclear peak.  If it can be avoided 
I do not wish to have to prepare by hand (which is what I have been 
doing for the last 7 years).
I have so far tested varying chopping times, the amount of tissue and 
micron size. 
Does anyone have any suggestions on how to obtain good results with 
the medimachine or has tried it on plant tissue?
Please any suggestions would be appreciated,
