Reponses indicated that there is some ambiguity in the question:
"Are methods established to ensure that immunoglobulin staining is instrinsic
and not extrinsic (cytophilic)?"

Most considered it an issue relating to staining to detect surface Ig light
chains in lymphoma immunophenotyping. One respondent had spoken with a member
of the checklist committee, who seemed to indicate that this was the correct
context.

To that end, strategies included:
*thorough washing to "elute" autologous Ig
*37C pre-incubation with calf, mouse or rabbit serum to block cytophilic
binding
*boolean gating strategies using CD45/SSC and CD19/SSC bitmaps
*ensuring that the sum of kappa and lambda does not exceed unity

There were several respondents who thought the question dealt "in general" with
delineating surface from cytoplasmic staining when using antibodies directed
against an epitope detectable on/in both sites.

Thank you all for your assistance.

Marc
