Dear list members We experienced a very unusual phenomenon as follows; A few days ago, a healthy donor was performed the peripheral blood progenitor cell collection on COBE Spectra following the G-CSF mobilization protocol for allogeneic stem cell transplantation to his brother suffering from an acute leukemia. Because one of two HLA haplotypes was not totally mismatched, the physician decided to select the CD34 positive stem cells and to reduce the T lymphocytes using CEPRATE system. After the procedure, CD34 positive cell count and lymphocyte subset tests were requested. Everthing had been fine as we expected. However, the proportion of CD19 positive lymphocytes in the final product was unbelievable. IT WAS OVER 90 PERCENT OF TOTAL LYMPHOCYTES. Even more, CD3 positive lymphocytes also expressed CD19. The lymphocyte subset profile of pre-CD34 selection procedure (PBPC product) was similar to that of normal peripheral blood and CD3 positive lymphocytes did not express CD19. We thought that it might be an error but the exact same phenomenon occurred on the next day. Here is the illustration of our results. CD19 | ... | . CD3- CD19+ 92% | ..... | ...
