Using an orienting flow nozzle and staining for DNA content using
Hoechst 33342 is the only reliable way to sort viable sexed sperm.
While the idea of using oligos makes sense, the problem is that you
have to kill the sperm to get the oligos into the DNA. No one to my
knowledge has been able to come up with any specific surface markers
that could be used to distinguish X from Y sperm without having to
fix the sperm.
