The recent thread about cord blood should prove useful to you, personally I
dedicate one color for the purpose of identifying the cells of interest, eg
CD45 or CD3, gating on fluorescence v. side scatter and using the other two
or three channels for subtyping these cells. This may not work for
neoplastic cells however.
