Apoptosis

I would like to broaden the current apoptosis debate by suggesting that
the flow community is getting too hung up over the question of how best
to tell whether a cell is dead or not.

Apoptosis as the final readout of a series of molecular events that can
result from cell siganlling or cell injury. I have become very impressed with
the power of multiparameter flow cytometry to map this sequence using
probes that sense alterations in cellular redox balance or calcium ion
regulation. This is where you start to see the effects of molecular
mechanisms that can influence whether or not a cell undergoes apoptosis,
such as increases in bcl-2 expression or p53 alterations.

Once you start to look at the upstream effectors you appreciate that a cell
is history long before it hits a positive TdT assay. And the reagents are
much cheaper!
