The best positive control of apoptosis are HL-60 cells incubated with
0.15 uM camtothecin for 3 - 4 h. Under these conditions all S phase
cells undergo apoptopsis (positive control) whereas G1 and G2 cells
do not (negative control in the same sample). We usually prepare a
batch of such cells which after 15 min fixation in 1% formaldehyde at
0 degrees is then stored in ethanol at -20 degrees C and lasts for
months as a control in our laboratory. Phoenix Flow Systems
commercially provides such a control. The attemts by us to use the
nucleases treated cells failed to give reproducible results.
