RE: Dyes for cell tracking experiments...

Hi,
At one stage we had a researcher here testing many dyes for cell tracking
and at that time BCECF was found to be the "best" (Nick King's
C.Parish/Weston recommendation). Since that time another carboxy fluorescein
CFSE seems to have been more favoured and it can work particularly well for
tracking cells which are dividing, class switching, when used in conjuction
with appropriate MoAb's.
One caveat on the PKH-26 is that in our hands it is toxic to sheep
lymphoid cells (I know the original post was mouse tumor). This has also
been the experience at at least one other site working on sheep cells here
in Oz. and after much trial and error it can be useful however if the same
experiment is done using CFSE the time course can be continued for a longer
time peroid with more cell circulating.
