sheath fluid preservative

Here, at the (University of Pennsylvania Cancer Center) Flow Cytometry
and Cell Sorter Shared Resource, we use a commercially available sheath
fluid (Stephens Scientific Diluent 2, distributed by Baxter/VWR). Below
is a listing of their ingredients...

NaCl (<1%)
di-water
2-phenoxyethanol
sodium phosphate dibasic
potassium phosphate monobasic

It is my understanding that the 2-phenoxyethanol has bacteriocidal
properties (Merck Index). I have seen very few instances where the
preservative has somehow gotten in the way of performing flow cytometric
analysis. ONE EXCEPTION HOWEVER WAS IN SORTING...ON THE BD
FACSORT...when comparing the results of a sort using sterile 1x PBS vs.
the commercially available sheath fluid (a la preservative), I noticed
that there was a more significant portion of dead cells when sorting with
the phenoxyethanol laced fluid.

Hope this helps....
