Lysosomes by flow

An excellent lysosomal probe is acridine orange. Incubation of live
cells with low concentration of AO (1 ug/ml) results in accumulation
of the dye in lysosomes which fluoresce red (>600 nm). All other cell
constituents (RNA and DNA) fluoresce green at this low AO
concentration. Stainability of lysosomes with AO is lost following
cell fixation or rupture of the cell plasma membrane and in late
phase of apoptosis. The method is described in Meth. Cell Biology Vol
41, pp. 185- 194, 1994.
