Hello

I have been comparing different lysing agents and was wondering if
it was possible and/or advisable to add Paraformaldehyde to directly
stained whole blood from HIV+ patients subsequently lysed using B-D
Autolyse if one used this reagent on the bench instead of in the Facs
prep workstation. The final volume is 2ml and I was considering
adding 200ul of a 10% Paraformaldehyde.

Could anybody please comment as the price per test would be very
competitive to Immunoprep and I think I'm right in that these are
the only 'no-spin' lysing reagents available. Another thing, is
Paraformaldehyde the best fixative for HIV positive samples, is it
better than Formalin and if so, why?

Thanks

Jane
