I run a FACStar+ here at the University of Massachusetts. Our policy for the
past six years has been not to sort samples stained with acridine orange
based on advice from BD. A researcher has a cell preparation stained with AO
which he would very much like to sort. His preliminary data was collected on
an Epics Profile. By diligently cleaning with detergent and bleach he is
confident that he removes residual AO from his system. He verifies this by
running unlabeled cells and watching to see if their fluorescence increases
due to picking up AO stuck in the tubing. He is trying to convince me that I
would be able to clean up the sorter this way.
I'm looking for advice - should we continue to adhere to our 'no AO' policy
or with the proper cleaning is this unwarranted?
