About calcium measurement

To all of you:

I would like to ask some questions about Ca2+ measurement.

First, we do not have UV excitation, so we cannot use Indo-1. We try to use
Fluo-3. In our studies, the cells were incubated in a temp- and pH
controlled chamber and we hope to measure the Ca2+ up to 90 mins, i.e. to
take the cells into the flow every 5 minutes for measurement. We wonder
the dye is suitable for such long time incubation at 37C. Is there anyone
who has such experience?

Second, is there anyone who combine Fura-red and Fluo-3 for ratio
measurement? we have the problem in generating a calibration curve. Can
someone tell me how to make this calibration experiment (e.g. incubation
time, temp., concentration, etc.)
